yHSC 人源化小鼠
人源CD34 +造血干细胞的M-NSG小鼠可以重建多谱系的人类免疫细胞,是进行肿瘤免疫和传染病研究的有效模型。
CD34+人源化小鼠系将人的脐带血、胎肝造血干细胞(HSC)移植到辐照清髓的M-NSG小鼠体内的模型。HSC移植后可以定植在小鼠骨髓,并不断产生各类造血或免疫细胞,如T细胞、B 细胞、NK 细胞、髓系细胞等。由于其免疫细胞是在小鼠体内发育而出,对小鼠宿主产生耐受,因而不会出现GvHD现象,人源免疫细胞将稳定存在,且重建后的免疫细胞类型较PBMC人源化小鼠丰富。但该类模型因为没有人T细胞发育所必需的人的胸腺,因此T细胞功能相对较弱。另外,该模型的髓系细胞发育不足,NK细胞含量相对较低,原因是某些调控这两类细胞发育的细胞因子在人鼠之间存在种属差异。
